Der junge Dr. Shaun Murphy ist ein sehr guter Chirurg und hat eine brillante Intuition in medizinischen Fragen. Da er ein Autist mit Inselbegabung ist, sind seine sozialen Fähigkeiten allerdings eingeschränkt. Als er eine Stelle in der Pädiatrie eines Krankenhauses im kalifornischen San José annimmt, sind die Verantwortlichen dort kritisch ihm gegenüber: Die an einer Immunschwäche leidende Patientin Tara will keine Genersatztherapie beginnen, weil sie sich als abstoßende Außenseiterin empfindet.
